233 A novel ex vivo skin culture device enables clinically representative skin testing

چکیده
Dermatology has the poorest rate of successful bench-to-clinic transitions. In part this is due to safety testing being historically performed on unrepresentative animal models, or replacement tests that use animal, rather than human data as benchmark for efficacy. As a result, preclinical models are unable faithfully replicate responses. Here, we examined whether novel environmentally relevant skin culture device able better mimic responses permeation and irritation. We engineered biphasic fluidic combining regulation atmospheric air, humidity, temperature, pH media flow, replicating physiological environment skin. Using device, assessed caffeine ibuprofen in ex vivo at baseline cultured five days found no significant difference absorption profiles (p 0.05 ANOVA with Dunnet’s multiple comparisons test). Additionally, irritation potential compounds, developing methodology based approved OECD 439 test. Approved validation chemicals, compounds incorrectly categorised were assessed. The chemicals correctly (100% accuracy), results fully replicated published clinical data. conclusion, physiologically was essential maintaining function clinically examples, demonstrate should replace traditional methods development accurate, human-representative analysis confident

Comparison of interleukin 10 homologs on dermal wound healing using a novel human skin ex vivo organ culture model.
BACKGROUND Anti-inflammatory cytokine interleukin (IL)-10 has been shown to induce regenerative healing in postnatal wounds. A viral homolog of IL-10 produced by human cytomegalovirus (CMV IL-10) similarly generates potent immunoregulatory effects, but its effects on wound healing have not been investigated. Currently, there are limited cost-effective methods of screening vulnerary therapeutics...
متن کاملA Novel Method for Skin Lesion Segmentation
Skin cancer has been the most usual and illustrates 50% of all new cancers detected each year. If they detected at an early stage, treatment can become simple and economically. Accurate skin lesion segmentation is important in automated early skin cancer detection and diagnosis systems. The aim of this study is to provide an effective approach to detect the skin lesion border on a purposed imag...

متن کاملDevelopment, standardization and testing of a bacterial wound infection model based on ex vivo human skin
Current research on wound infections is primarily conducted on animal models, which limits direct transferability of these studies to humans. Some of these limitations can be overcome by using-otherwise discarded-skin from cosmetic surgeries. Superficial wounds are induced in fresh ex vivo skin, followed by intradermal injection of Pseudomonas aeruginosa under the wound. Subsequently, the infec...
متن کاملEx vivo confocal microscopy: a diagnostic tool for skin malignancies.
Confocal microscopy is an imaging tool that uses a laser system to image the skin noninvasively with cellular resolution. With both in vivo and ex vivo systems, tissue can be imaged rapidly without damaging the sample. Herein, we discuss the capabilities of the ex vivo confocal microscope and its implementation for treatment and management of cutaneous malignancies.
